Untersuchung von Multikomponentenreaktionen zur Darstellung unnatürlicher Glycinderivate

  • Mit der Blickrichtung auf die Synthese und Darstellung neuer Verbindungen zur Verbesserung der Lebensqualität, Grundbedürfnisse und Gesundheit einer modernen Gesellschaft sind neue Strategien, welche die Konzepte der Nachhaltigkeit, Effizienz und Umweltschutz verinnerlichen, von immenser Bedeutung. Multikomponentenreaktionen (MCRs) offerieren hierfür einen attraktiven Lösungsansatz. Als Eintopfreaktionen zeichnen sich MCRs durch hohe Stufen- sowie Atomökonomien aus und können ferner dazu beitragen Kosten, Zeit, Energie und Abfälle einzusparen. Durch den modularen Charakter von MCRs lassen sich darüber hinaus schnell und effizient eine Vielzahl unterschiedlicher Molekülstrukturen generieren. Vor diesem Hintergrund konnten im Rahmen dieser Forschungsarbeit einige neue, kombinatorische Konzepte zur Synthese α-substituierter Glycinderivate entwickelt werden. Ausgehend von einer weiterentwickelten, Pd-katalysierten, enantioselektiven 3-Komponentenreaktion auf Basis von Sulfonsäureamiden, Glyoxylsäure Monohydrat und Boronsäuren konnte eine unerwartete Hintergrundreaktion festgestellt werden, die sich schlussendlich als erste katalysatorunabhängige Petasis-Reaktion auf Basis von Sulfonsäureamiden als Amin-Komponente darstellte. In Anbetracht der Aufgabe eine ressourcensparende und nachhaltigere Aufbaumöglichkeit von unnatürlichen Aminosäurederivaten zu entwickeln, gelang es im Zuge dieser Doktorarbeit weiterhin eine Pd-katalysierte, decarboxylative 3-Komponentenreaktion aufzubauen und zu etablieren. Dabei bieten Arylcarbonsäuren als Schlüsselkomponenten in einem decarboxylativen Konzept mit Palladium eine kostengünstigere und umweltfreundlichere Alternative zu den bisher bestehenden Methoden mit Organoboronsäuren oder anderer Organometall-Spezies. Überraschenderweise konnte im Zuge der Untersuchungen zur Substratbreite der Aldehyd-Komponente eine direkte, decarboxylative Addition zwischen den eingesetzten Arylcarbonsäuren und den verschiedenen Glyoxylsäureestern beobachtet werden. Dabei eröffnete diese Methode in ersten Untersuchungen eine interessante Möglichkeit zur Darstellung entsprechender Mandelsäurederivate.
  • With a view towards synthesis and presentation of novel molecules to improve the quality of life, basic needs and health issues of a modern society, new strategies that internalize concepts of sustainability, efficiency and environmental protection are of outranging importance. Multi-component reactions (MCRs) offer an attractive solution for this. As a one-pot reaction, MCRs are characterized by high step and nuclear economies and can also help to save costs, time, energy, and waste. Due to their modular nature, many different molecular structures can be generated very fast and effectively. Some new, combinatorial concepts for the synthesis of α-substituted glycine derivatives could be developed within the framework of this research work. Based on the development of a Pd-catalyzed, enantioselective 3-component reaction with sulfonamides, glyoxylic acid monohydrate and boronic acids, an unexpected background reaction could be determined, which ultimately presented itself as the first catalyst-independent petasis reaction based on sulfonamides as an amine component. In view of the task to develop a resource-saving and more sustainable route for unnatural amino acid derivatives, it was possible to further establish a Pd-catalyzed, decarboxylative 3-component reaction within this doctoral thesis. Aryl carboxylic acids as key components in a decarboxylative concept with palladium offer a more cost-effective and environmentally friendly alternative towards existing methods with organoboronic acids or other organometallic species. Surprisingly, during this study on the substrate scope of the aldehyde component, a direct, decarboxylative addition between the aryl carboxylic acids and various glyoxylic acid esters could be observed. Initial investigations reveal that this method opens an interesting possibility for the representation of corresponding mandelic acid derivatives.
